Sermorelin (CAS: 86168-78-7) is a synthetic peptide consisting of the first 29 amino acids of the 44-amino acid human growth hormone-releasing hormone (GHRH). This N-terminal fragment retains the full biological activity of the native hormone, as the first 29 residues contain the complete receptor-binding domain. Sermorelin was among the earliest GHRH analogs studied and has an extensive research history spanning over four decades in growth hormone physiology.
Sermorelin binds to the GHRH receptor (GHRHR) on anterior pituitary somatotroph cells, activating adenylyl cyclase through Gs-protein coupling and increasing intracellular cAMP concentrations. This signaling cascade stimulates both the synthesis and secretion of growth hormone (GH) in a pulsatile, physiological manner. Unlike exogenous GH, sermorelin preserves the natural feedback mechanisms of the hypothalamic-pituitary-somatotroph axis, including sensitivity to somatostatin-mediated inhibition, maintaining physiological GH pulsatility.
