Ipamorelin (CAS: 170851-70-4) is a synthetic pentapeptide (Aib-His-D-2-Nal-D-Phe-Lys-NH2) belonging to the growth hormone secretagogue (GHS) class. It was developed as a highly selective agonist of the growth hormone secretagogue receptor (GHS-R1a, also known as the ghrelin receptor). Among the GHS peptide family, Ipamorelin is notable for its selectivity in stimulating GH release with minimal effects on adrenocorticotropic hormone (ACTH), cortisol, or prolactin secretion.
Ipamorelin binds to the GHS-R1a receptor on pituitary somatotrophs, triggering growth hormone release through phospholipase C activation and intracellular calcium mobilization. Unlike other growth hormone secretagogues such as GHRP-6 or hexarelin, ipamorelin demonstrates a narrow selectivity profile, stimulating GH release in a dose-dependent manner without appreciably altering cortisol, ACTH, or prolactin levels.
