Hyaluronic acid (HA) is a non-sulfated glycosaminoglycan built from repeating disaccharide units of glucuronic acid and N-acetylglucosamine. It is one of the most abundant extracellular matrix components in vertebrate tissues, with particularly high concentrations in synovial fluid, vitreous humor, and skin dermis. Unlike most other glycosaminoglycans, HA is not covalently attached to a protein core -- it exists as a free polysaccharide that can reach molecular weights of several million daltons in native tissue.
The biology of HA is surprisingly complicated for a compound that looks structurally simple. Molecular weight turns out to matter a lot: high-molecular-weight HA (above roughly 500 kDa) is generally considered anti-inflammatory and space-filling, while lower-molecular-weight fragments (below 500 kDa, and especially oligosaccharides below 20 kDa) are pro-inflammatory and angiogenic. This size-dependent bioactivity arises because different molecular weight ranges engage different receptor classes -- CD44 and RHAMM interact differently depending on chain length, and the toll-like receptors TLR2 and TLR4 respond primarily to low-molecular-weight fragments.
Research applications for injectable-grade HA include osteoarthritis models, intra-articular injection studies, tissue engineering scaffolds, fibroblast proliferation in cited models, and dermal biology. The research-grade material offered here allows controlled investigation without the lot-to-lot variability of tissue-derived HA. Some groups use it as a baseline matrix component in hydrogel systems for 3D cell culture, where its ability to engage CD44 on cells provides a relevant physiological context.

Store lyophilized powder at 2-8°C, protected from moisture. Reconstitute in sterile water or phosphate-buffered saline. High-molecular-weight HA dissolves slowly -- allow 24-48 hours with gentle agitation rather than vigorous vortexing, which can shear the polymer chains. Once reconstituted, store at 4°C and use within 7 days.
Size determines both physical properties (viscosity, space-filling capacity) and biological signaling. Native high-MW HA forms a viscoelastic matrix that is poorly inflammatory. When HA is degraded by hyaluronidases -- which happens in injured or inflamed tissue -- the resulting smaller fragments act as danger signals through TLR2/4 and promote inflammation. This means that the HA fragment size in your experimental system will shape the biology you observe. Always verify the molecular weight distribution of your batch before designing experiments where fragment size could confound results.
Cosmetic-grade HA is typically formulated with other ingredients, preservatives, and carriers, and endotoxin levels are not controlled to the same standard. Research-grade HA provides documented molecular weight, purity, and ideally an endotoxin specification -- critical for cell culture work where even low endotoxin levels (above ~0.1 EU/ml) can activate macrophages and complicate any inflammatory readout. For in vivo studies, pyrogen testing is a minimum requirement before use.
