Gonadorelin is a synthetic decapeptide identical to endogenous GnRH (gonadotropin-releasing hormone), the hypothalamic hormone studied for its role in driving LH and FSH secretion from the anterior pituitary in research models. It is the pharmacological reference standard for the GnRH pathway -- native sequence, no modifications, no analogues. Gonadorelin is used in research to probe pituitary responsiveness (the GnRH stimulation paradigm) and to model central GnRH signalling. Each vial contains 2mg.
The GnRH pathway operates on pulse frequency. In research models, pulsatile GnRH secretion is studied for its role in HPG-axis (LH/FSH) signalling, stimulating LH and FSH release, while continuous GnRH stimulation desensitizes GnRH receptors and is associated with suppression of gonadotropin secretion -- the mechanism exploited pharmacologically by GnRH agonist compounds studied in hormone-sensitive cancer and endometriosis research models. Gonadorelin's native sequence and short half-life (minutes) preserve this pulsatile-stimulatory behavior in research, making it distinct from long-acting GnRH agonist analogues like triptorelin or leuprolide that initially stimulate and then suppress through desensitization.
For research applications, gonadorelin is used to study the hypothalamic-pituitary-gonadal (HPG) axis, probe hypothalamic function, investigate GnRH receptor pharmacology, and model endocrine responses in reproductive biology research.

Gonadorelin is native GnRH -- the same sequence as the endogenous hormone. It has a short half-life (minutes) and is studied for stimulating LH and FSH in a pulsatile manner in research models. GnRH agonists like triptorelin, leuprolide, and goserelin are modified analogues with much longer half-lives. Paradoxically, continuous exposure to long-acting GnRH agonists desensitizes GnRH receptors, initially associated with a hormone surge followed by suppression of LH and FSH -- the mechanism studied in hormone-sensitive cancer and endometriosis research models. Gonadorelin's short half-life means it stimulates rather than suppresses when dosed intermittently in these models.
The GnRH stimulation paradigm (also called the GnRH or LHRH challenge) assesses pituitary responsiveness by measuring LH and FSH response to an injection of gonadorelin in research models. Samples are taken before and at 30 and 60 minutes after injection. A robust LH and FSH rise indicates intact pituitary responsiveness in the model; a blunted or absent response indicates reduced pituitary response. The paradigm is used in research to distinguish hypothalamic from pituitary contributions to the HPG axis -- helping determine whether the limiting factor is inadequate GnRH signal or inadequate pituitary response to that signal.
GnRH receptor signaling is frequency-encoded. In research models, the pituitary responds differently to different GnRH pulse frequencies: optimal pulsatility (roughly 1 pulse per 90-120 minutes) is associated with maintained LH and FSH secretion. Faster pulses shift the ratio toward LH; slower pulses favor FSH. Continuous stimulation desensitizes the receptor entirely and is associated with suppressed gonadotropin secretion. This frequency sensitivity is why pulsatile GnRH delivery is studied for restoring gonadotropin output in research models of hypothalamic hypogonadism, while continuous GnRH agonist exposure suppresses it -- opposite outcomes from the same receptor in these models.
