What is Dihexa?
Dihexa (N-hexanoic-Tyr-Ile-(6)-aminohexanoic amide) is a synthetic hexapeptide-derived angiotensin IV analog that was developed at Washington State University. Its mechanism is unusual in the nootropic research space: rather than targeting dopamine, serotonin, acetylcholine, or even the classical neurotrophic receptors like TrkB, Dihexa potentiates signaling through the hepatocyte growth factor (HGF) / c-Met receptor pathway. HGF/c-Met was known primarily as a growth and survival factor in peripheral tissues and tumor biology; research from the Bhargava and Bhattacharya labs at WSU identified that this pathway also drives synaptogenesis — the formation of new synaptic connections — in the hippocampus.
The potency figure that gets cited is striking: the original preclinical papers reported Dihexa to be approximately 10 million times more potent than BDNF at promoting synaptic connectivity in hippocampal cell assays. That claim deserves some context — the comparison isn't straightforward because BDNF and Dihexa act through entirely different receptor systems. But the functional synaptogenesis data in cognitive models (including scopolamine-impaired spatial memory tests in rodents) is notable. At 20mg the compound has enough quantity for extended in vitro studies or multiple animal experiments. The HGF/c-Met pathway in CNS remains substantially undercharacterized relative to BDNF/NGF, making Dihexa a useful tool for researchers interested in this space.

What is the HGF/c-Met pathway and why is it relevant to the CNS?
HGF (hepatocyte growth factor) and its receptor c-Met are widely known in oncology for their roles in cancer cell invasion and survival. In the CNS, the HGF/c-Met signaling axis has more recently been recognized as a regulator of neuronal survival, axon growth, and synaptic connectivity — particularly in the hippocampus. c-Met is expressed on hippocampal neurons, and HGF/c-Met signaling promotes synaptogenesis and dendritic spine formation through downstream activation of PI3K/AKT and MAPK/ERK pathways. Dihexa was identified by screening for compounds that could potentiate this pathway with high potency, which the WSU group achieved by starting from the angiotensin IV/AT4 receptor framework.
