The DAC stands for Drug Affinity Complex — a maleimidopropionate linker on a Lys extension that reacts with the free thiol of albumin's Cys-34 residue in circulation. Once bound, the peptide stays in circulation for as long as albumin does: several days. This is meaningfully different from CJC-1295 without DAC (which has amino acid substitutions for DPP-4 resistance but still clears in hours) and from native GHRH, which has a plasma half-life of minutes.
The Teichman 2006 Phase I/II study in healthy adults showed single-dose CJC-1295 DAC produced sustained GH and IGF-1 increases that lasted six days or more, with a clear dose-response relationship. That's the published clinical benchmark. For research purposes, the extended pharmacokinetics mean once-weekly dosing can maintain elevated GH axis activity throughout the protocol — relevant to body composition studies, IGF-1 research, and any investigation where prolonged GH elevation is the experimental variable rather than an acute pulse.
At the receptor level, CJC-1295 DAC activates GHRH receptors on anterior pituitary somatotrophs through the same cAMP-dependent mechanism as native GHRH. The DAC modification doesn't change receptor binding kinetics or downstream signaling — only how long the compound remains available to bind. One important caveat: the claim in early literature that CJC-1295 DAC maintains natural pulsatile GH release despite continuous albumin-bound peptide presence has been questioned. Sustained GHRH stimulation produces a different GH secretion pattern than the natural intermittent hypothalamic pulse. Researchers designing studies should account for this: CJC-1295 DAC produces sustained GH axis stimulation, not physiological pulses. For pulsatile GH research, CJC-1295 without DAC is the appropriate tool.
