Mechano Growth Factor (MGF) is an alternatively spliced variant of the IGF-1 gene expressed preferentially in skeletal muscle in response to mechanical loading or injury. The C-terminal E-peptide sequence unique to MGF is responsible for its distinctive biological activity -- specifically its ability to activate muscle satellite cells, the resident stem cells of skeletal muscle. Each vial contains 2mg of the native (non-PEGylated) MGF peptide.
The key thing to understand about native MGF is its instability in physiological fluids. Native MGF has a plasma half-life measured in minutes, which means it is degraded rapidly after systemic administration. This isn't necessarily a problem for all research designs -- local intramuscular injection delivers the peptide directly to the target tissue where satellite cells reside, bypassing the stability problem. For research focused on local muscle biology, native MGF at the injection site has the advantage of physiological route of action and no PEG modification to complicate interpretation.
The contrast between native MGF and PEG-MGF represents a meaningful research design choice. Native MGF better reflects what happens in the muscle microenvironment after mechanical loading. PEG-MGF allows for systemic administration and longer exposure times. Both approaches have their place depending on the research question.

Store at -20°C before reconstitution. Due to its instability, native MGF reconstituted solutions should be used quickly -- within 7 days at 2-8°C. Minimize freeze-thaw cycles. For studies where longer stable supply is needed, PEG-MGF is available as an alternative.
Use native MGF for local intramuscular injection studies where the goal is to study satellite cell activation in the injected muscle tissue -- this mirrors the physiological context of mechanical load-induced MGF expression. The short half-life is less of a problem when delivering directly to target tissue. Use PEG-MGF for systemic administration studies where sustained plasma concentrations are needed, or for studies where once-daily or less frequent dosing is required. PEG-MGF better suits experiments looking at whole-body effects or cross-muscle signaling.
Native MGF has a plasma half-life of approximately a few minutes. After systemic (IV or SC) injection, it is cleared very rapidly by proteolytic degradation and renal filtration. In the local muscle microenvironment after intramuscular injection, the effective exposure time is longer because the peptide is partially protected from circulating proteases, but this is still measured in hours rather than days. This instability is the main limitation of native MGF for systemic research applications and the reason PEGylated forms were developed.
Satellite cells normally exist in a quiescent state beneath the muscle fiber basal lamina. MGF E-peptide binding activates these cells, causing them to proliferate (producing a larger pool of myogenic progenitors) and then differentiate -- fusing with existing muscle fibers to add new myonuclei, or forming entirely new fibers. This process is essential for muscle repair after injury and for hypertrophy in response to training load. MGF acts upstream of IGF-1 signaling in this cascade, with IGF-1 taking over the differentiation phase after satellite cells have been activated by MGF.
