What is DSIP?
Delta Sleep-Inducing Peptide is a nine-amino-acid peptide first pulled from the cerebral venous blood of rabbits during slow-wave sleep in 1977. The name is slightly misleading — DSIP does not simply knock you out. It modulates sleep architecture, specifically promoting delta wave (stage 3/4 NREM) activity without suppressing REM. That is a very different thing from sedation.
CAS: 62568-57-4. A neuromodulatory peptide that sits at an unusual intersection: sleep science, stress physiology, and pain research, all in one compound.
What makes it odd
DSIP does not fit neatly into any single category. It modulates sleep, but also reduces stress-induced ACTH and cortisol. It has analgesic properties through endogenous opioid facilitation. It affects thermoregulation. And it stimulates LH release, which is a completely different axis. Most peptides do one thing well. DSIP touches five different systems, and nobody has fully explained how a nonapeptide manages that.
Mechanism
Promotes delta wave sleep without suppressing REM — it changes sleep quality, not just sleep quantity. Reduces ACTH and cortisol under stress. Facilitates endogenous opioid activity for analgesic effects. Modulates hypothalamic temperature regulation. Stimulates LH through an unclear pituitary mechanism. The multi-system profile suggests DSIP works upstream of these individual effects, possibly at the level of hypothalamic coordination.
