How to Reconstitute Peptides: Step-by-Step Protocol

Before You Start

Reconstitution is the process of dissolving lyophilized (freeze-dried) peptide powder into a liquid solution for research use. Done correctly, it takes about 10 minutes. Done incorrectly, you can damage your peptide irreversibly. This guide covers the technique that preserves peptide integrity.

Equipment Checklist

  • Bacteriostatic water (0.9% benzyl alcohol — provides antimicrobial protection for multi-use)
  • Insulin syringes (1mL, 29-31 gauge)
  • Alcohol swabs (70% isopropanol)
  • Clean workspace (ideally a laminar flow hood, but a sanitized bench surface works)
  • Nitrile gloves

Step 1: Sanitize

Swab the rubber stoppers on both vials — the peptide vial and the bacteriostatic water vial — with alcohol swabs. Let air dry completely (30-60 seconds). This prevents microbial contamination.

Step 2: Draw Bacteriostatic Water

Using an insulin syringe, draw your calculated volume of bacteriostatic water. Common volumes depend on desired concentration — use our Reconstitution Calculator for exact calculations.

Critical Technique

In the next step, you must add water along the inside wall of the vial, letting it flow down gently. Never inject directly onto the lyophilized powder — the force and localized hydration can denature the peptide and create insoluble aggregates.

Step 3: Add Water to Peptide

Insert the needle at an angle through the rubber stopper. Position the needle tip against the inside wall of the vial. Slowly depress the plunger, allowing the water to trickle down the glass wall and pool at the bottom. The lyophilized cake will begin dissolving on contact.

Step 4: Gentle Dissolution

Do not shake the vial. Instead:

  • Let the vial sit for 2-3 minutes after adding water
  • Gently roll the vial between your palms if the peptide hasn't fully dissolved
  • For stubborn peptides, swirl gently in a circular motion
  • Most peptides dissolve completely within 5-10 minutes

The solution should be clear and colorless (or blue-green for GHK-Cu). Cloudiness or particles indicate a problem — do not use.

Step 5: Label & Store

Mark the vial with: peptide name, concentration (mg/mL), date reconstituted, and volume added. Store at 2-8°C (standard refrigerator). Use within 4 weeks for optimal activity.

Common Mistakes to Avoid

  • Injecting water directly onto the powder (causes aggregation)
  • Shaking vigorously (denatures peptide structure)
  • Using non-sterile water (contamination risk)
  • Forgetting to swab stoppers (bacterial contamination)
  • Leaving reconstituted peptide at room temperature (accelerated degradation)
