N-Acetyl Epitalon Amidate is a chemically modified form of Epitalon, the AEDG tetrapeptide first isolated from the bovine pineal gland by Vladimir Khavinson's group in the 1980s. The N-terminal acetylation and C-terminal amidation are a standard pharmaceutical trick for improving peptide stability -- both modifications protect against the exopeptidases that would otherwise cleave the termini rapidly. The result is a compound with similar putative activity to Epitalon but better resistance to enzymatic degradation in biological media.
Epitalon's research profile centers on telomerase activation and its downstream effects on telomere length. The original Khavinson studies, conducted in cell culture and animal models, reported lengthening of telomeres in somatic cells following Epitalon exposure, an effect attributed to upregulation of hTERT (telomerase reverse transcriptase). This has since been reproduced in some experimental systems, though the magnitude and durability of the effect vary. Neuroendocrine regulation is another area of interest -- the peptide appears to influence melatonin secretion by the pineal gland, which has downstream effects on circadian regulation and antioxidant status.
The N-Acetyl Amidate form is typically used when longer experimental windows are needed or when repeated dosing in animal studies would otherwise require very high quantities of the standard peptide to overcome rapid degradation. Whether the modifications alter receptor binding relative to native Epitalon is not well characterized in the published literature -- this is a reasonable area of uncertainty that active researchers should account for in their experimental design.

Store at -20°C. The modified termini improve stability relative to native Epitalon, but lyophilized powder should still be kept away from humidity. Once reconstituted in sterile water or bacteriostatic water, store at 4°C and use within 30 days. Avoid freeze-thaw cycling of reconstituted solution.
The core AEDG sequence is identical. The acetyl group added to the N-terminus and the amide group added to the C-terminus block the free amine and carboxyl that exopeptidases prefer to attack. In practice this extends serum half-life in animal studies. The trade-off is that the modifications slightly change the peptide's charge and potentially its interaction with any endogenous receptor -- something worth considering when comparing results directly to native Epitalon studies.
Telomere length is commonly measured by quantitative PCR relative to a reference gene (T/S ratio method) or by Southern blot of terminal restriction fragments. Telomerase activity itself is assessed with the TRAP (telomeric repeat amplification protocol) assay. hTERT mRNA levels by RT-PCR serve as a proxy for transcriptional upregulation. Each method has its own sources of variance, so convergent evidence across multiple assays strengthens any claim about telomere biology.
