Every GLP-1 receptor agonist in clinical development is trying to be a better version of this compound. Native GLP-1 (7-36)-amide is the endogenous incretin hormone secreted by intestinal L-cells after nutrient ingestion — thirty amino acids, plasma half-life of 1-2 minutes. DPP-4 cleaves the His-Ala dipeptide at the N-terminus almost immediately after secretion. CAS: 89750-14-1. MW 3,297.7 Da. That rapid degradation was the problem that drove the entire field toward engineered analogs — liraglutide, semaglutide, tirzepatide — all designed to survive longer than the native ligand. Native GLP-1 is the reference: what all of them mimic, what all of them are benchmarked against.
The short half-life, usually a liability in compound development, is an asset in certain research contexts. Studying acute GLP-1R activation dynamics, receptor binding kinetics with unmodified pharmacology, DPP-4 degradation pathway analysis, or genuine post-prandial incretin physiology requires the native ligand. Analogs with week-long half-lives can't produce that data.
The receptor pharmacology is incretin biology in its original form: GLP-1R binding activates adenylyl cyclase, raising cAMP in pancreatic beta cells. The resulting insulin secretion is glucose-dependent — insulin only releases during hyperglycemia, not at baseline. That selectivity is what makes the incretin effect protective, and it's best studied with the compound the body actually produces. Acute receptor activation dynamics, kinetic binding studies, DPP-4 degradation pathway research, and post-prandial incretin physiology all require the unmodified native ligand. Long-acting analogs like semaglutide can't replicate the acute signaling profile that the body's own post-meal GLP-1 produces.
The incretin effect refers to the observation that oral glucose produces more insulin secretion than the same amount of IV glucose — because eating triggers GLP-1 (and GIP) release from the gut, which amplifies pancreatic beta cell response. The glucose-dependence of GLP-1's insulin-stimulating effect means insulin only rises when plasma glucose (per cited trials) is elevated, not at baseline.
