PEGylated Mechano Growth Factor is a modified form of MGF (a splice variant of IGF-1) stabilized by polyethylene glycol conjugation. The PEG modification extends the half-life substantially -- native MGF has a very short plasma half-life (minutes to hours) that limits its utility in systemic research contexts. PEGylation typically extends this to 24+ hours, enabling once-daily or even less frequent dosing in animal models. Each vial contains 2mg.
MGF (Mechano Growth Factor) is derived from the IGF-1 gene through alternative splicing. When muscle is subjected to mechanical load, this splice variant is preferentially expressed over systemic IGF-1. The MGF E-peptide (the C-terminal extension unique to this splice form) is responsible for most of its distinct biology -- particularly its role in activating muscle satellite cells (stem cells resident in skeletal muscle). Satellite cell activation is the initiating step in muscle repair after injury. Native MGF's extreme instability in physiological fluids is the primary reason PEGylated forms are used for systemic administration research.
The biological question being addressed with PEG-MGF is whether systemic delivery of a stabilized MGF form can reproduce the local satellite cell activation that normally occurs in response to mechanical loading -- and whether this has applications in muscle wasting, aging, or injury recovery research.
PEG-MGF is used in research on satellite cell biology, muscle regeneration and repair, muscle wasting (sarcopenia, cachexia), IGF-1 splice variant pharmacology, and the role of mechanical loading signals in muscle adaptation.

Native MGF has a plasma half-life of minutes, which makes it impractical for systemic administration research. The peptide is rapidly degraded before it can produce measurable effects at distant tissues. PEGylation -- attachment of polyethylene glycol chains -- shields the peptide from proteolytic degradation and renal clearance, extending half-life to 24+ hours. This allows meaningful plasma concentrations to be maintained with manageable dosing frequency in animal models. Without PEGylation, most MGF research uses local injection directly into target muscle tissue.
MGF and systemic IGF-1 are both encoded by the IGF-1 gene but differ in how they are spliced. When skeletal muscle experiences mechanical load or damage, alternative splicing produces MGF with a unique C-terminal E-peptide sequence. This E-peptide is not present in systemic IGF-1 and is responsible for MGF's satellite cell-activating properties. Systemic IGF-1 promotes muscle protein synthesis and has anabolic effects, but MGF's role in satellite cell activation for repair is mechanistically distinct and not replicated by IGF-1.
Satellite cells are muscle stem cells that reside quiescent beneath the basal lamina of muscle fibers. When muscle is damaged or overloaded, they activate, proliferate, and fuse with existing or new fibers to repair and hypertrophy the muscle. This is the primary mechanism of post-injury regeneration. Declining satellite cell activity with aging (sarcopenia) and in muscle wasting conditions is a research focus for understanding why muscle repair becomes less effective. MGF's role in activating these cells makes it a research tool for studying satellite cell biology and potential interventions for muscle wasting.
